Spurs are also front-runners to sign Vestergaard's £25m-rated international team-mate Joachim Andersen from Lyon.
He spent last season on loan at Fulham, catching Spurs' eye, but new manager Nuno Espirito Santo is set to make the final call on whether to sanction his signing.
Spurs remain in talks to reach an agreement with Bologna over defender Takehiro Tomiyasu too.
He can play in two positions the north Londoners want to strengthen, right back and centre-back and new managing director Fabio Paritici is especially keen to bring Tomiyasu to Spurs.
Personal terms have been agreed with Tomiyasu, who has been included in Japan's Olympic squad, though the two teams are still slightly apart on his valuation.
Spurs had a £12.9m bid rejected and are reluctant to pay more than £15.5m while Bologna are holding out for £17.2m.
