The Agonist – zu interview “Days Before the World Cry” • metal.de

Riser released his EP “Days Before the World Cry” in October. This strong release was reason enough for us to beat the band, who were kind enough to answer our questions via email. Congratulations on your new EP, which in my opinion is one of your strongest to date. Why...

A particulate saponin/TLR agonist vaccine adjuvant alters lymph flow and modulates adaptive immunity

Adjuvants can enhance immune responses against poorly immunogenic antigens, making them an important component of many vaccines despite the limited number currently approved for clinical use. Silva et al. developed a new adjuvant formed through self-assembly of saponin with the Toll-like receptor 4 agonist monophosphoryl lipid A (MPLA) into nanoparticle structures. Compared with a panel of tested adjuvants including other saponin-based formulations, saponin/MPLA nanoparticles (SMNP) induced superior antibody and germinal center responses after immunization with a poorly immunogenic HIV antigen in both mice and rhesus macaques. SMNP increased lymphatic flow in a mast cell–dependent manner and antigen delivery to draining lymph nodes, providing insight into the mechanisms by which saponin adjuvants act to enhance humoral immune responses.

Underrepresentation of diverse populations and clinical characterization in opioid agonist treatment research: A systematic review of the neurocognitive effects of buprenorphine and methadone treatment

J Subst Abuse Treat. 2021 Oct 23:108644. doi: 10.1016/j.jsat.2021.108644. Online ahead of print. INTRODUCTION: The relative neurocognitive effects of the two most common opioid agonist treatments (OAT; buprenorphine and methadone) for opioid use disorder (OUD) are poorly understood. The aim of this systematic review is to examine the neurocognitive effects of OAT (buprenorphine and methadone) and the clinical and sociodemographic characteristics of study samples.

Type 2 Diabetes: SGLT2 Inhibitors and GLP-1 Receptor Agonists

Jennifer B. Green, MD: There’s a role for all these medications. As you know, most people with diabetes will progress over time and need additional glucose-lowering therapy. We’re fortunate that we have a large number of tools to use. Where it can become tricky is figuring out not so much how to design care for the person with newly diagnosed diabetes—that’s still reasonably straightforward—but how we incorporate these drugs into an already complex diabetes treatment regimen. This is where the diabetes care specialist can be helpful.

The Agonist: Hope In A Weeping World

It’s an almost unwritten fact at this point that most bands – where they could – have spent the last eighteen months beavering away on new material or something new and exciting for fans. It’s been as much out of necessity to tide them over in an elongated album cycle as it has been borne of bands simply having much more time on their hands. Canadian melodic death metallers THE AGONIST are certainly no different, having spent a good portion of 2021 working on their latest EP, Days Before The World Wept. It’s a few weeks prior to release when we catch up with their vocalist Vicky Psarakis who, it transpires, has been even busier than most.

Client resistance to hepatitis C treatment initiation in opioid agonist treatment clinics in Sydney, Australia: A qualitative study

Drug Alcohol Rev. 2021 Nov 28. doi: 10.1111/dar.13414. Online ahead of print. INTRODUCTION: Opioid agonist treatment (OAT) clinics play a key role in achieving elimination of hepatitis C virus (HCV) globally. Previous research has identified barriers to HCV treatment uptake in OAT clinics; however, most studies were conducted prior to the introduction of direct-acting antiviral treatments (DAA). It remains unclear whether progress has been made in responding to barriers and what challenges persist in this setting.

Biased beta-agonists may provide better control of asthma, other obstructive lung diseases, drug discovery study shows

Beta-agonists (β-agonists) are the only drugs that directly open narrowed airways and make it easier to breathe for millions of people with asthma, a chronic respiratory disease. These inhaled medications activate the b2-adrenergic receptors (β2AR) on airway smooth muscle cells and relax them, dilating airways and increasing air flow. However,...

Sosei, Neurocrine ink $2.6B neuropsychiatric collaboration

LONDON – Sosei Heptares is to get $100 million up front in a potential $2.6 billion deal with Neurocrine Biosciences Inc., in which the pair will develop muscarinic receptor agonists in the treatment of schizophrenia, dementia and other neuropsychiatric disorders. San Diego-based Neurocrine is taking rights to a portfolio of small molecules against specific subtypes of the muscarinic receptor family, targeting M1 and M4 alone, plus a dual M1/M4 agonist.

“This is the first beta-agonists ever known to relax airway smooth muscle and treat asthma without any detectable tachyphylaxis” – says study author

For millions of patients suffering from asthma, a chronic respiratory disease, beta-agonists (-agonists) are the only medications that directly expand restricted airways and make breathing easier. These inhaled medicines activate and relax the b2-adrenergic receptors (2AR) on airway smooth muscle cells, dilating airways and boosting air flow. However, for a...

Zealand Pharma Announces Successful Outcome of Phase 1b Clinical Trial With GLP1-GLP2 Dual Receptor Agonist

Zealand Pharma A/S recently announced topline results from its Phase 1b multiple ascending dose trial with dapiglutide. In this randomized, double-blind, placebo-controlled, multiple ascending dose trial, four cohorts of 10 healthy subjects were allocated to 4 different dose levels, randomized to either 4 weekly doses of dapiglutide or placebo. The objective of the trial was to evaluate the safety and tolerability of multiple doses of dapiglutide. Additionally, the pharmacokinectics and pharmacodynamics of dapiglutide were assessed during this trial.
